By car it normally takes around 3 hours / 3 hours and a half, without any stop and the cost for the highway is €36,40. As you probably know is not possible to arrive in Venice by car.
Actually You can park your car in Piazzale Roma choosing between different parkings open all day. Otherwise there is the Tronchetto parking based on the island of Tronchetto. Then you can get to Venice's old city centre using private or public transportation.
In Piazzale Roma you will find the following parking places: Autorimessa Comunale, Garage S.Marco and S.Andrea (open). From here you can go to Venice by foot or by waterbus.
In Tronchetto island you will find a very large car park. From here you can go to Venice only with the waterbus line 82.
Alternatively you can leave the car outside Venice, in Mestre (normally it is easier and cheaper) but it takes longer to reach the city center.
If you are planning to visit Florence and/or Venice I really suggest you to book ahead
your hotel and visits to Museums, guided tours, gondola rides, etc...
